Aim: The present study was conducted to detect the presence of canine parvovirus (CPV) among diarrheic dogs in Himachal Pradesh and to identify the most prevalent antigenic variant of CPV based on molecular typing and sequence analysis of VP2 gene.
Materials and Methods: A total of 102 fecal samples were collected from clinical cases of diarrhea or hemorrhagic gastroenteritis from CPV vaccinated or non-vaccinated dogs. Samples were tested using CPV-specific polymerase chain reaction (PCR) targeting VP2 gene, multiplex PCR for detection of CPV-2a and CPV-2b antigenic variants, and a PCR for the detection of CPV-2c. CPV-2b isolate was cultured on Madin-Darby canine kidney (MDCK) cell lines and sequenced using VP2 structural protein gene. Multiple alignment and phylogenetic analysis was done using ClustalW and MEGA6 and inferred using the Neighbor-Joining method.
Results: No sample was found positive for the original CPV strain usually present in the vaccine. However, about 50% (52 out of 102) of the samples were found to be positive with CPV-2ab PCR assay that detects newer variants of CPV circulating in the field. In addition, multiplex PCR assay that identifies both CPV-2ab and CPV-2b revealed that CPV-2b was the major antigenic variant present in the affected dogs. A PCR positive isolate of CPV-2b was adapted to grow in MDCK cells and produced characteristic cytopathic effect after 5thpassage. Multiple sequence alignment of VP2 structural gene of CPV-2b isolate (Accession number HG004610) used in the study was found to be similar to other sequenced isolates in NCBI sequence database and showed 98-99% homology.
Conclusion: This study reports the first detection of CPV-2b in dogs with hemorrhagic gastroenteritis in Himachal Pradesh and absence of other antigenic types of CPV. Further, CPV-specific PCR assay can be used for rapid confirmation of circulating virus strains under field conditions.

Aim: This investigation is placed in the context of continuity of a preliminary study already published; it was conducted in cattle, sheep, goats, horses, and dogs; the main aim is to reveal and develop criteria for the animal species determination based on the morphometric parameters of red blood cells.
Materials and Methods: Blood samples were taken from the jugular vein; and the smears were confectioned on slides immediately after the blood collection and stained according to the May-Gründwald Giemsa method. For the morphometric study, three parameters were considered which are: The diameter, the circumference, and the surface of erythrocytes; and measurements were achieved using the OPTIKATM Vision Pro software. Statistical analysis was performed by both analysis of variances and Student’s t analytical tests.
Results: The recorded data showed that the three morphometric parameters of red blood cells are higher in dogs followed, respectively, by those of horses, cattle, and sheep, whereas, the goats have the lowest ones. In addition, the obtained results allowed us to propose new reference values for the circumference and the surface of erythrocyte in considered species.
Conclusion: This investigation permit concluding that from a drop of blood it is possible to characterize the different animal species, taking into account the diameter, the circumference, and the surface of erythrocytes.

Background: The One Health (OH) approach, which seeks to bring together human and animal health, is particularly suited to the effective management of zoonotic diseases across both sectors. To overcome professional silos, OH needs to be taught at the undergraduate level. Here, we describe a problem-based learning activity using the OH approach that was conducted outdoors for 3rd-year veterinary students in Malaysia.
Materials and Methods: A total of 118 students, divided into two groups, completed the activity which spanned 1½ days at a deer park adjacent to a wilderness area. Students were asked to evaluate the activity using an online survey that had quantitative and qualitative components.
Results: Response rate was 69.5%. The activity was rated excellent by 69.5% and good by 30.4%. Levels of satisfaction were high on a range of criteria. 97.5% of students intended to take action in their studies as a result of what they had learned.
Conclusions: Delivery of an outdoor problem-based learning activity using OH approach was very successful in terms of participation, knowledge delivery and understanding, and the willingness of students to integrate OH into their future practice. For the improvement of future programs, the involvement of other disciplines (such as Medical, Biology, Biotechnology, Biomedical, and Public Health) is being considered.

Background and Aim: There are global public health and economic concerns on chemical residues in food of animal origin. The use of antibiotics in dairy cattle for the treatment of diseases such as mastitis has contributed to the presence of residues in dairy products. Penicillin residues as low as 1 ppb can lead to allergic reactions and shift of resistance patterns in microbial population as well as interfere with the processing of several dairy products. Antibiotic monitoring is an essential quality control measure in safe milk production. This study was aimed at determining antibiotic residue contamination and the level of penicillin in dairy products from Fulani cattle herds in Oyo State.
Materials and Methods: The presence of antibiotic residues in 328 samples of fresh milk, 180 local cheese (wara), and 90 fermented milk (nono) from Southwest, Nigeria were determined using Premi® test kit (R-Biopharm AG, Germany) followed by high-performance liquid chromatography analysis of penicillin-G residue.
Results: Antibiotic residues were obtained in 40.8%, 24.4% and 62.3% fresh milk, wara and nono, respectively. Penicillin-G residue was also detected in 41.1% fresh milk, 40.2% nono and 24.4% wara at mean concentrations of 15.22±0.61, 8.24±0.50 and 7.6±0.60 μg/L with 39.3%, 36.7% and 21.1%, respectively, containing penicillin residue above recommended Codex maximum residue limit (MRL) of 5 μg/L in dairy. There was no significant difference between the mean penicillin residues in all the dairy products in this study.
Conclusion: The results are of food safety concern since the bulk of the samples and substantial quantities of dairy products in Oyo state contained violative levels of antibiotic residues including penicillin residues in concentrations above the MRL. This could be due to indiscriminate and unregulated administration of antibiotics to dairy cattle. Regulatory control of antibiotic use, rapid screening of milk and dairy farmers’ extension education on alternatives to antibiotic prophylaxis, veterinary prescriptions and withdrawal periods are recommended to prevent residues.

Aim: To investigate the status, problems and prospects of Japanese quail (Coturnix coturnix japonica) farming in selected areas of Bangladesh.
Materials and Methods: The study was conducted in 14 districts of Bangladesh, viz., Dhaka, Narayanganj, Munshiganj, Mymensingh, Netrakona, Faridpur, Jessore, Khulna, Satkhira, Kushtia, Bogra, Naogaon, Comilla, and Sylhet during the period from July 2011 to June 2012. A total of 52 quail farmers were interviewed for data collection using a structured questionnaire. Focus group discussions were also carried out with unsuccessful farmers and those want to start quail farming. Workers of quail farms, quail feeds and medicine suppliers, quail eggs and meat sellers were also interviewed regarding the issue.
Results: Out of 52 farms, 86.5% were operated by male, 67.3% farmers did not receive any training and 92.3% farmers had no earlier experience of quail farming although 58.0% farmers primary occupation was quail farming. Most of the farms (63.4%) were mixed in type having ≤5000 birds of two or three varieties. About 80.7% farms were operated separately round the year with no other poultry and 83.0% farmers wanted to expand their farming. The average pullet weight 14.5±0.12, 11.0±0.07, 12.0±0.22, and 12.8±0.17 g; age at the first lay 46.0±0.04, 42.0±0.31, 42.0±0.09, and 45.2±0.05 days; rearing period 15.0±0.01, 12.0±0.14, 15.0±0.32, and 15.2±0.18 months; culling period 15.5±0.14, 13.0±0.06, 15.0±0.03, and 15.4±0.26 months were for layer, parent stock, hatchery, and mixed farms, respectively. Most of the layer farms had an average egg production of ≤5000/day and net profit BDT 0.75/egg. However, an average number of birds, hatchability and net profit per day-old-chick were ≤5000, 76.8% and BDT 2.75, respectively, in the hatchery. Broiler quails were sold at 30 days with mean weight of 110.8 g and net profit BDT 9.02/bird. The major constraints of quail farming were higher feed price, outbreak of endemic diseases, lack of proper knowledge, farmers training, proper market access, difficulties of parent stock collection, inadequate biosecurity practices, and limited access to veterinary care. Thus, a proper training on quail farming, bio-security management, and government subsidy on feeds could make quail farming sustainable in Bangladesh.
Conclusions: The study concludes that Japanese quail farming has enormous potentiality and could be an alternative to chicken farming particularly in providing gainful employment, supplementary income and as a valuable source of meat and egg, quail farming should be encouraged and promoted in Bangladesh.

Aim: The interpretation of conventional polymerase chain reaction (PCR) assay results is often limited to either positive or negative (non-detectable). The more robust quantitative PCR (qPCR) method is mostly reserved for quantitation studies and not a readily accessible technology in laboratories across developing nations. The aim of this study was to evaluate a semi-quantitative method for conventional PCR amplicons using digital image analysis of electrophoretic gel. The potential applications are also discussed.
Materials and Methods: This study describes standard conditions for the digital image analysis of PCR amplicons using the freely available ImageJ software and confirmed using the qPCR assay.
Results and Conclusion: Comparison of ImageJ analysis of PCR-electrophoresis gel and qPCR methods showed similar trends in theFusobacterium necrophorum DNA concentration associated with healthy and periodontal disease infected wallabies (p≤0.03). Based on these empirical data, this study adds descriptive attributes (“more” or “less”) to the interpretation of conventional PCR results. The potential applications in low-income veterinary laboratories are suggested, and guidelines for the adoption of the method are also highlighted.

Aim: To evaluate the protective and ameliorative effect of aqueous sea buckthorn leaf extract (SLE) on hemato-biochemical profile in lead intoxicated Wistar rats.
Materials and Methods: An experiment was conducted for 60 days. 36 adult male Wistar rats with a mean body weight of 177.8±12.6 g were divided into five groups and were subjected to various daily oral treatment regimens. Group I served as a negative control receiving only feed and water, Group II (positive control for lead) received lead acetate at 250 ppm in drinking water, and Group III (positive control for SLE) received SLE at 100 mg/kg b.wt. Animals in Group IV received a combination of lead acetate at 250 ppm in drinking water for the first 45 days and SLE at 100 mg/kg b.wt. throughout the experimental period of 60-day, and in Group V for the last 15 days of the trial after the administration of lead acetate until the first 45 days of the trial to study the protective and ameliorating effects of SLE, respectively. Blood samples were collected from retro-orbital fossa of each rat on 0th, 45th, and 60th day of the experiment for hemato-biochemical analysis including hemoglobin (Hb), packed cell volume (PCV), serum total protein, albumin, globulin, albumin:globulin ratio, cholesterol, urea, and creatinine.
Results: Significantly (p<0.01) lower levels of serum total proteins and albumin, and a significantly (p<0.01) higher serum cholesterol, urea and creatinine levels were observed in Group II (lead intoxicated group) in comparison to Group I (negative control). Administration of SLE at 100 mg/kg body wt. to lead intoxicated Wistar rats resulted in normalization of almost all the biochemical parameters studied in both the treatment Groups, i.e., IV and V (protective and ameliorative). However, the effects were more pronounced in the protective group. No effects of SLE supplementation were observed on Hb levels. PCV levels improved in protective groups, but no effect was observed in ameliorative group in comparison to lead intoxicated groups.
Conclusion: SLE administration at 100 mg/kg b.wt. to lead intoxicated Wistar rats may be used to protect/ameliorate lead induced biochemical alterations in Wistar rats.
