ABSTRACT

Background and Aim: The emergence of methicillin-resistant Staphylococcus aureus (MRSA) as a highly pathogenic strain in veterinary and human medicine is a growing global problem. This study aimed to evaluate MRSA isolates of human and animal origin against various antibiotics in Yogyakarta, Indonesia.

Materials and Methods: The susceptibility test was carried out by the disk diffusion method using Mueller-Hinton agar against nine antibiotic disks. Methicillin-resistant S. aureus strains were genetically confirmed through mecA gene detection encoding for methicillin resistance by polymerase chain reaction.

Results: All 240 S. aureus strains isolated from animals and humans were resistant to penicillin G (P) (100% and 99%, respectively), followed by ampicillin (AMP), amoxicillin (AML), oxacillin (OX), erythromycin (E), clindamycin (DA), tetracycline (TE), gentamicin (GEN), and ciprofloxacin (CIP). Eighty-three MRSA strains were resistant to OX (100%), P (100%), AMP (99.27%), AML (95.52%), E (87.77%), TE (71.33%), DA (63.24%), GEN (38.81%), and CIP (26.87%).

Conclusion: The antimicrobial resistance pattern of S. aureus human isolates was similar to their animal counterpart, with 77.20% of MRSA strains classified as multidrug-resistant (MDR) bacteria. These findings indicate an increase in MDR S. aureus strains of animal origin in Yogyakarta, thus raising public health concerns about MRSA zoonotic spread.

Background and Aim: Given the rise in stray and imported dogs in Egypt over the past 5 years, it is surprising that no report of Brucella canis infection in dogs or humans has been documented in Egypt's published papers. This study aimed to detect the presence of antibodies against the rough (B. canis) and smooth Brucellae among dogs in Egypt and to characterize the Brucella species circulating in dogs.

Materials and Methods: Blood samples (n = 449) were collected from owned and stray dogs in the Greater Cairo region (n = 309) and Damietta governorate (n = 140). The apparent, true, and total seroprevalence of canine brucellosis caused by B. canis infection were calculated using the 2-mercaptoethanol tube agglutination test (2-ME TAT) and rapid slide agglutination test (RSAT). We used the rose Bengal test (RBT) and the buffered acidified plate antigen test (BAPAT) to check the serum samples from dogs for the presence of antibodies against smooth Brucellae. Three polymerase chain reaction (PCR) assays - Bruce-ladder PCR, B. canis species-specific PCR (BcSS-PCR), and Abortus Melitensis Ovis Suis (AMOS)-PCR - were used to determine the Brucella species in the buffy coats of the serologically positive dogs.

Results: The overall apparent and true prevalence of B. canis infection in dogs were estimated to be 3.8% and 13.2%. The estimated true prevalence in stray dogs (15%) was higher than in owned dogs (12.5%). The BAPAT and the RBT using smooth antigens revealed that 11 (2.4%) and 9 (2%) were positive. Bruce-ladder PCR targeting eryCABC, and Polysaccharide deacetylase genes was able to identify B. canis in nine out of 17 buffy coat samples. AMOS-PCR identified the eight undetermined Brucella species by Bruce-ladder PCR as Brucella abortus (n = 4) and Brucella melitensis (n = 4). To exclude the presence of Brucella suis, a one-step species-specific BcSS-PCR was performed and specifically amplified all B. canis DNA (n = 9) the same as did the Bruce-ladder PCR.

Conclusion: To the best of our knowledge, this is the first report of B. canis detection in dogs in Egypt. Molecular identification of B. abortus and B. melitensis in the Egyptian canines highlights the role of stray dogs in brucellosis remerging in Brucellosis-free dairy farms. Brucella canis infection can be diagnosed specifically with the one-step BcSS-PCR. The obtained results set-an-alarm to the veterinary authorities to launch plans to control this disease in dogs.

ABSTRACT

Background and Aim: Campylobacteriosis causes gastrointestinal tract lesions in adults and children and may result in severe complications. The primary sources of infection are infected animals and animal products. Immunochemical methods effectively diagnose intestinal infections but require highly specific antigens to detect their antibodies. This study aimed to obtain two recombinant immunogenic antigens of Campylobacter jejuni, an outer membrane protein with a molecular weight of 18 kDa (Omp18) and the major outer membrane protein (MOMP) with a molecular weight of 45 kDa, and evaluate their suitability for the serological diagnosis of campylobacteriosis using immunochromatographic assay (ICA).

Materials and Methods: The C. jejuni Omp18 and MOMP gene sequences were synthesized de novo (Macrogen, Korea) and cloned into the pET32 expression plasmid. Using these genetic constructs, electrocompetent cells of the Escherichia coli BL21 strain were transformed and cultured under various conditions. Antigens were purified and refolded using metal affinity chromatography. The properties of the purified proteins were studied by western blotting, liquid chromatography with tandem mass spectrometry, and enzyme-linked immunosorbent assay (ELISA).

Results: We developed two recombinant E. coli BL21 cells producing rOmp18 and Recombinant MOMP (rMOMP) antigens with molecular weights of 36 and 64 kDa, respectively. Amino acid sequence analysis of the obtained antigens showed complete homology with the reference sequences in the PubMed NCBI database. Western blotting using positive-control sera demonstrated the specificity of the recombinant antigens. The results of ELISA with 94 bovine sera showed the interaction of recombinant antigens with specific antibodies.

Conclusion: The obtained rOmp18 and rMOMP antigens can detect antibodies in the serum of infected or recovered animals and can be used to develop ICA.

ABSTRACT

Background and Aim: Ipomoea aquatica (Water Spinach) is the most potential for livestock growth performance, including chickens, pigs, cattle, and goats, especially in a tropical country like Cambodia. It is not only an alternative feed source but also one kind of supplemented feed for goat raising. Supplementation with Water Spinach in the utilization of low-quality tree foliage results in an increase in dry matter intake in goat production. This study aimed to identify the effectiveness of supplementation of Water Spinach in the utilization of Mimosa pigra and Leucaena leucocephala leaf in in vitro fermentation.

Materials and Methods: The study was designed according to a 2 × 2 factorial arrangement in randomized design of seven treatments with different ratios consisted of different three types of dietary treatments, including M. pigraL. leucocephala, and Water Spinach. The treatments were arranged according to a completely randomized design and were as follow: T1 = M. pigra leaf (100%); T2 = L. leucocephala leaf (100%); T3 = M. pigra leaf and L. leucocephala leaf (50% and 50%); T4 = M. pigra leaf and Water Spinach (99.5% and 0.5%); T5 = L. leucocephala leaf and Water Spinach (99.5% and 0.5%); T6 = M. pigra leaf and Water Spinach (99% and 1%); and T7 = L. leucocephala leaf and Water Spinach (99% and 1%). A total of 200 mg (dry matter) of dietary treatments were prepared in a 60 mL syringe. Each treatment was replicated 3 time. Gas recording of each treatment lasted for 3 days. In vitro was performed for 72 h, was followed by Makkar method. Gas production was recorded at 2, 4, 8, 12, 24, 36, 48, and 72 h of incubation by using strict anaerobic technique. A mixture of rumen fluid and dietary treatments were carried out under continuous flushing with CO2 in sharking incubator at 39°C. After incubating for 72 h, the ammonia concentration (NH3-N) was measured and recorded to identify pH, nutrient digestibility, and ammonia concentration (NH3-N).

Results: Nutrient digestibility of the treatment with Water Spinach supplement in the utilization of L. leucocephala was obtained at a higher digestibility than treatment with M. pigra (p < 0.05). Gas production was different between groups (p < 0.05). Treatment with only M. pigra leaf had the highest gas production (A), while treatment with Water Spinach supplementation had the lowest gas production (A). At 0–24 h, the treatment with L. leucocephala leaf and Water Spinach 0.5% had the highest gas production, but after 24 h, M. pigra leaf and Water Spinach 1% and L. leucocephala leaf and Water Spinach 0.5% produced more gas compared to the other treatments (p < 0.05).

Conclusion: The supplementation of Water Spinach 1% in treatment with M. pigra and L. leucocephala leaf resulted in increased degradability, gas production, and NH3-N concentration without a change in the pH value rumen condition. Based on these results, it is recommended that the level of Water Spinach supplementation should be 1% of dietary intake. Future studies should consider investigating the rumen ecology associated with Water Spinach supplementation. Feeding with Water Spinach remains a good supplement for ruminant performance; therefore, further studies should be conducted using Water Spinach in ruminant feeding in both metabolic and feeding trials.

Background and Aim: Public health and food safety are gaining attention globally. Consumer health can be protected from chemical residues in meat by early detection or screening for antibiotic residues before selling the meat commercially. However, conventional practices are normally applied after slaughtering, which leads to massive business losses. This study aimed to use portable surface-enhanced Raman spectroscopy (SERS) equipped with multivariate curve resolution-alternation least squares (MCR-ALS) to determine the concentrations of enrofloxacin, oxytetracycline, and neomycin concentrations. This approach can overcome the problems of business loss, costs, and time-consumption, and limit of detection (LOD).

Materials and Methods: Aqueous solutions of three standard antibiotics (enrofloxacin, oxytetracycline, and neomycin) with different concentrations were prepared, and the LOD for each antibiotic solution was determined using SERS. Extracted pig urine was spiked with enrofloxacin at concentrations of 10, 20, 50, 100, and 10,000 ppm. These solutions were investigated using SERS and MCR-ALS analysis. Urine samples from pigs at 1 and 7 days after enrofloxacin administration were collected and investigated using SERS and MCR-ALS to differentiate the urinary enrofloxacin concentrations.

Results: The LOD of enrofloxacin, oxytetracycline, and neomycin in aqueous solutions were 0.5, 2.0, and 100 ppm, respectively. Analysis of enrofloxacin spiking in pig urine samples demonstrated the different concentrations of enrofloxacin at 10, 20, 50, 100, and 10,000 ppm. The LOD of spiking enrofloxacin was 10 ppm, which was 10 times lower than the regulated value. This technique was validated for the first time using urine collected on days 1 and 7 after enrofloxacin administration. The results revealed a higher concentration of enrofloxacin on day 7 than on day 1 due to consecutive administrations. The observed concentration of enrofloxacin was closely correlated with its circulation time and metabolism in pigs.

Conclusion: A combination of SERS sensing platform and MCR-ALS is a promising technique for on-farming screening. This platform can increase the efficiency of antibiotic detection in pig urine at lower costs and time. Expansion and fine adjustments of the Raman dataset may be required for individual farms to achieve higher sensitivity.

Background and Aim: The Goliath chicken is a slow-growing chicken with a high slaughter weight but whose carcass characteristics and meat quality have not yet been documented. This study aimed to evaluate the effect of age at slaughter and sex on the carcass characteristics and technological and sensory meat quality of Goliath chickens raised in southern Benin.

Materials and Methods: Data on the carcass characteristics and technological and sensory quality of meat were collected from 80 chickens raised in confinement and divided into two groups. The first group was reared for 12 weeks of age and the second group was for 20 weeks of age. The animals were individually weighed using an electronic balance (Terraillon 5000g) and then slaughtered. The different parts of the chicken carcass was weighed using the same method.

Results: The live, hot carcass, cold carcass, wing, thigh, and tail weights of males were significantly greater than those of females (p < 0.01). The wishbone muscle and thigh and tail assembly pH was lower in the 20-week-old chickens than in the 12-week-old chickens. The brightness and red index of the wishbone of the 20-week-old chickens were significantly higher than those of the 12-week-old chickens (p < 0.001). The red index of the wishbone and the yellow index of the thigh and tail assembly of females were lower than those of males.

Conclusion: The meat of 20-week-old Goliath chickens was juicier than that of 12-week-old chickens. Thus, the carcass composition of Goliath chickens is better at 20 weeks of age, especially in males.

ABSTRACT

Background and Aim: Blastocystis hominis is an intestinal protozoan in humans and animals. The parasite causes mild-to-severe intestinal complications, such as diarrhea, in healthy humans and immunocompromised hosts. This study aimed to determine the antiprotozoal activity of Boesenbergia rotunda (L.) Mansf and Ganoderma lucidum (Fr.) Kart extracts against B. hominis.

Materials and Methods: Antiprotozoal activity of B. rotunda and G. lucidum extracts against B. hominis subtype 3 was determined using the erythrosin B exclusion assay, confirmed by a time-kill study. The morphology of the parasite treated with the extracts was observed by a scanning electron microscope. The phytochemicals present in B. rotunda and G. lucidum extracts were identified by gas chromatography-mass spectrometry analysis.

Results: Both B. rotunda and G. lucidum extracts demonstrated strong antiprotozoal activity with similar minimum inhibitory concentration (MIC) values of 62.5 μg/mL. At 4× MIC and 8× MIC, both B. rotunda and G. lucidum extracts, and metronidazole inhibited the growth of B. hominis by up to 90% after 12 h treatment. Blastocystis hominis cells treated with B. rotunda extract, G. lucidum extract, and metronidazole were deformed and withered when compared with the control. Geraniol and versalide were found as the main compounds in B. rotunda and G. lucidum extracts, respectively.

Conclusion: These results indicate the potential medicinal benefits of B. rotunda and G. lucidum extracts in the growth inhibition of B. hominis.

ABSTRACT

Background and Aim: Streptococcosis is a common bacterial disease in red tilapia, in which Enterococcus faecalis infection has not been widely reported. This study aimed to evaluate the efficacy of pellets that contain chicken E. faecalis-induced immunoglobulin Y (IgY) to treat and prevent streptococcosis in red tilapia.

Materials and Methods: We conducted a 28-day study for immunoprophylaxis and immunotherapy, each using four groups with two replications: Healthy control fish (KS), non-IgY pellets (PA and TA), pellets with 25% egg yolk containing E. faecalis-induced IgY (PB and TB), and pellets with 50% egg yolk containing E. faecalis-induced IgY(PC and TC). Indirect enzyme-linked immunosorbent assay was performed on prototype pellets produced with an IgY suspension at 1.63 mg/mL as the standard optical density curve. For the immunoprophylaxis study, pellets of 3% of the average body weight of the experimental fish (0.50 g per fish per day) were given daily until day 14 before the challenge test with E. faecalis (2.1 × 109 Colony-forming unit/mL peroral) on day 15. The data from the observation period on days 15–28 were analyzed. For the immunotherapy study, pellets of 3% of the average body weight (0.50 g per fish per day) were given daily for 21 days (days 8–28) 7 day spost-infection. The data from the immunotherapy study were collected during the observation period on days 8–28. Statistical analysis was performed on non-specific immune variables: Total leukocytes, monocytes, lymphocytes, neutrophils, phagocytic activity, and macrophage capacity; and the semi-quantitative distribution of melanomacrophage centers (MMCs) in the lymphoid organs, such as spleen and liver. Photomacrographic data were analyzed descriptively and qualitatively by comparing the healing process and clinical signs found between experiments in the immunotherapy study.

Results: The pellet with 50% egg yolk with an IgY at 2.43 mg/g pellet, 3% of body weight once daily, was the best formula on experimental fish. The administration of this formulation can also increase non-specific immunity and the distribution of MMCs in the spleen and liver with a survival rate of 55% for 14 days of challenge period in the immunoprophylaxis study and 70% for 21 days of therapy period in the immunotherapy study.

Conclusion: Immunoglobulin Y can be a prophylactic and therapeutic agent against streptococcal infections caused E. faecalis in red tilapia with an optimum dosage of 2.43 mg/g pellet.

ABSTRACT

Background and Aim: Antimicrobial resistance, especially antibiotic resistance, is one of the most severe public health challenges. Antibiotic resistance occurs when bacteria avoid and fight the mechanism of action of antibiotic drugs. This study aimed to determine the resistance of Escherichia coli from the milk of Ettawa crossbreed dairy goat at Blitar Regency, East Java, Indonesia, with the antibiotics streptomycin, sulfonamides, and trimethoprim.

Materials and Methods: A total of 34 milk samples of Ettawa crossbreed dairy goats were used in this study. The initial stages of this research included tests of the physical properties, isolation, and identification of E. coli. Then, the E. coli isolates were tested for antibiotic resistance using the Kirby–Bauer method.

Results: The results showed that all samples were positive for E. coli. The physical properties of milk, namely, color, odor, flavor, and consistency, were normal. The results of the alcohol test showed normal acidity, and the specific gravity of goat milk met the criteria, with an average specific gravity of 1.0295 g/mL. The results of the antibiotic resistance test showed that 4 (12%) samples were resistant to streptomycin, 5 (15%) to sulfonamide, and 3% to trimethoprim.

Conclusion: The prevalence of E. coli from Ettawa crossbreed dairy goats in Blitar Regency, East Java, Indonesia, was 100%. Furthermore, this E. coli isolate exhibited resistance to antibiotics streptomycin, sulfonamides, and trimethoprim. The use of antibiotics in the dairy goat industry in Indonesia should be controlled to prevent the spread of resistant E. coli from animals to humans through the food chain and prevent the emergence of multidrug-resistant E. coli.

ABSTRACT

Background and Aim: Bacterial and viral infections affect the welfare of animals and lead to large economic losses in dairy cattle breeding due to decreased productive indicators and increased culling rates. In modern dairy farming, farmers are looking for effective solutions to prevent and minimize infectious disease risks. To this end, the most relevant study field is the search for gene sites that impact production and health. This study aimed to determine the nature of the distribution of the relative frequencies of alleles and genotypes of polymorphic prolactin (PRL) and nitric oxide synthase (NOS2) in Holstein cows and identify the relationship of these genes with resistance to mastitis and bovine leukemia.

Materials and Methods: For this study, we chose cows because infectious diseases affect the amount of lactation and milk quality. Holstein cattle with mastitis and bovine leukemia were selected. Animal genotypes were determined by restriction fragment length polymorphism (RFLP) of polymerase chain reaction (PCR) products. The results were analyzed using a nonparametric statistical method using Microsoft Excel 2010 and Statistica 6.0.

Results: In healthy animals, 94 genotypes were identified for both genes under study. For bPRLbPRL-RsaIAA (72) was the most common genotype and bPRL-RsaIBB (4) the least; for NOS2bNOS2-HinfIAB (47) was the most common genotype and bNOS2-HinfIAA the least (21). In animals with leukemia, 34 genotypes were identified. For PRLbPRL-RsaIAA (25) was the most common genotype and bPRL-RsaIBB (2) the least; for NOS2bNOS2-HinfIBB (17) was the most common genotype and bNOS2-HinfIAA (3) the least. In animals with mastitis, 67 genotypes were identified. For PRLbPRL-RsaIAA (43) was the most common genotype and bPRL-RsaIBB (6) the least; for NOS2bNOS2-HinfIBB (31) was the most common genotype and bNOS2-HinfIAA (7) the least. The distribution of genotypes of polymorphic bPRL and bNOS2 generally coincides, and bPRL-RsaIBB is the most common genotype. In groups of sick animals, the number of bNOS2-HinfIAA homozygotes was lower than that of the control group. In particular, the proportion of animals with the bNOS2-HinfIAA genotype with bovine leukemia was 8.7% and with mastitis was 10.3% compared with 22.4% in healthy animals. These data support the possible association of the bNOS2-HinfIAA genotype with resistance to infection. The frequency of the bPRL-RsaIB allele was higher in groups of sick animals. This allele is associated with increased milk productivity, suggesting that highly productive animals are less resistant to the incidence of viral bovine leukemia and mastitis of bacterial etiology.

Conclusion: DNA amplification of Holstein cattle for the polymorphic regions of PRL and NOS2 using the PCR-RFLP method revealed a possible connection between the distribution of relative allele frequencies of bPRL and bNOS2 and resistance to viral and bacterial infections. Thus, in groups of sick animals, the frequency of bPRL-RsaIBB, associated with increased milk production compared with the theoretically calculated equilibrium value was higher and the number of homozygotes bNOS2-HinfIAA was lower than in the control group. In conclusion, animals with increased milk production were more prone to diseases, such as mastitis and bovine leukemia.

ABSTRACT

Background and Aim: Anemia is an important factor in surviving chronic kidney disease (CKD). Anemia in CKD is associated with various factors, such as inadequate production of erythropoietin and the availability of iron and its binding protein. Reduced total iron-binding capacity (TIBC) and iron concentrations may be related to their urinary loss along with proteinuria. This study aimed to determine the urinary loss of iron and transferrin (TF) in relation to the degree of proteinuria.

Materials and Methods: The study was performed on 37 dogs with CKD. Dogs were divided according to the severity of proteinuria into two groups based on the mean of urinary protein–creatinine (UPC) ratio into UPC ratio <4 and UPC ratio >4. The hematocrit (HCT), blood chemistries, plasma iron, plasma TF, UPC ratio, urinary iron per creatinine ratio (U-Iron/ CR), and urinary TF per creatinine ratio (U-TF/CR) were evaluated.

Results: Anemia was associated with the severity of renal impairment as demonstrated by reduction of HCT when staging of CKD was higher. Dogs with UPC ratio >4 had higher urinary loss of both U-Iron/CR (p < 0.01) and U-TF/CR (p < 0.001) with lower plasma TIBC (p < 0.001). The UPC ratio was positively correlated with both U-Iron/CR (r = 0.710, p < 0.001) and U-TF/CR (r = 0.730, p < 0.001) but negatively with TIBC (r = –0.462, p < 0.01).

Conclusion: Proteinuria was associated with urinary loss of both iron and TF which may contribute to anemia in CKD.

ABSTRACT

Background and Aim: Animal trypanosomiasis is a major contributor to agricultural and economic losses, especially in sub-Saharan Africa. We have shown that some animal species expressed genes that are significant players in immune response to bovine trypanosomosis, impeding signs and symptoms of the disease. We hypothesize that such animals are contributors to disease transmission dynamics and severe outcomes. Therefore, this study aims to ascertain trypanosome species diversity in cattle and their potential role as reservoirs for the transmission of human disease.

Materials and Methods: We performed a molecular genotyping of trypanosome internal transcribed spacer 1 (ITS-1) and 18S ribosomal RNA genes on genomic DNA extracts from randomly sampled N'Dama cattle from slaughterhouses in Nigeria. We identified trypanosome species circulating among the animals through polymerase chain reaction and genomic sequencing. We performed multiple sequence alignments as well as conducted a phylogenetic relationship between identified species.

Results: In all, 9 of 127 (7.1%) samples were positively amplified (band sizes ranging from 250 bp to 710 bp), including an isolate with two distinct bands (700 and 710 bp), indicating two trypanosome types. Sequence similarity and homology analysis identified four species, namely: Trypanosoma vivaxTrypanosoma congolense forest type, T. congolense savannah type, and Trypanosoma brucei. Interestingly, one of the bands, additionally verified by nucleotide sequencing, was identified as a human trypanosome (Trypanosoma brucei gambiense), confirming our hypothesis that cattle are potential reservoir hosts for human trypanosomes.

Conclusion: Overall, we observed different trypanosome species in our study area, with animals on the same farm infected with multiple species, which could complicate treatment and disease control strategies. Finding human trypanosome species strengthens the argument that disease transmission dynamics are modulated by other vertebrates, further complicating control programs.

ABSTRACT

Background and Aim: Using simulators in high education enables practical training by repetition in circumstances close to reality reducing the stress of both animal and operator. The limited resources of veterinary schools, the increase in the number of students in lecture halls, and the low availability of animals for teaching due to welfare regulations, reduce teaching opportunities with live animals being simulator as the better alternative. This study aimed to assess the efficacy of a low-cost handmade simulator (SIMCA-COW) in eight veterinary students inexperienced in palpation, ultrasonography evaluation, and artificial insemination in cows.

Materials and Methods: Two sets of exercises were carried out: structure localization by rectal palpation and ultrasonography were evaluated by the inexpert veterinary students in the SIMCA-COW simulator. Also, evaluated the length of time to insert the insemination catheter through the cervix in the inert simulator during four sessions.

Results: All the students were able to palpate both structures and to locate the body and both uterine horns by ultrasonography. Cervix and follicles were located by 5/8 (62.5%) students and 3/8 (37.5%) students found the corpus luteum by ultrasonography. A reduction in time span between the first and fourth intrauterine insemination attempts was observed (8.26 ± 2.7 vs. 3.69 ± 1.7; mean ± standard error; p < 0.05).

Conclusion: The simulator validated in this study (SIMCA-COW) allows training and learning by repetition, saving the limitations found in live animal practice.

ABSTRACT

Background and Aim: Keratoconjunctivitis sicca (KCS) is predominantly an immune-mediated chronic inflammatory ocular disease that is commonly diagnosed in dogs. This study aimed to compare the conventional use of topical immunosuppressant tacrolimus 0.03% eye drops and a new therapy injectable homologous platelet-rich plasma (HPRP) into the third eyelid gland and inferior and superior palpebral conjunctiva of dogs with KCS.

Materials and Methods: A total of 66 eyes from 33 dogs were evaluated. The eyes were divided into three equal groups: Negative control group, tacrolimus group (TG), and homologous platelet-rich plasma group (HPRPG). The animals were evaluated using the Schirmer's tear test-1 (STT-1), osmolarity test (OT), strip meniscometry test (SMT), tear film break-up test (TBUT), fluorescein test, lissamine green test (LGT), and cytological and histopathological analyses.

Results: In TG, there was a significant increase (p < 0.05) in the STT-1 and SMT values, and goblet cell count in the palpebral conjunctiva by the end of the study. In HPRPG, 36% (four dogs) received three applications, 55% (six dogs) received two applications, and 9% (one dog) received one application before the initial ocular signs improved. There was a significant decrease (p < 0.05) in the lymphocyte and neutrophil counts of the palpebral conjunctiva in HPRPG than in TG. Both groups showed equivalent improvements in TBUT, OT, and LGT values.

Conclusion: Tacrolimus 0.03% eye drops were more efficient than HPRP in increasing tear production and the number of goblet cells. However, injectable HPRP was more efficient than tacrolimus in decreasing the number of conjunctival inflammatory cells. Treatment with injectable HPRP requires an average of two to three applications, is safe and feasible, and can be used as a cheaper alternative or as an adjuvant to conventional treatment with topical immunosuppressants.

Background and Aim: Brucellosis is an infectious and zoonotic disease that affects people's health and the economy in most countries. Brucellosis is still prevalent in several Indonesian regions. This study aimed to analyze the correlation between the characteristics, knowledge, attitudes, and practices (KAP) of dairy farmers in Bogor District in supporting brucellosis control and surveillance programs.

Materials and Methods: The study was cross-sectional. Data were collected through interviews with 151 dairy farmers in Bogor Regency, West Java, Indonesia. The outcome is brucellosis surveillance and control practice among dairy farmers, and the variables include individual characteristics, knowledge, and attitudes toward brucellosis surveillance and control. Descriptive analysis and path analysis were used in statistical analysis.

Results: The majority of farmers' knowledge, attitudes and practices were moderate, with the percentages 67.55%, 60.92%, and 41.72% respectively. Formal education, training, and dairy rising length are variables that have a direct and significant impact on knowledge level. Knowledge is the variable that influences the overall level of attitude. Age, knowledge, and attitude are factors that influence the practice of brucellosis surveillance and control.

Conclusion: Although the practice level of brucellosis surveillance and control for dairy farmers in Bogor Regency is moderate, efforts to improve it are still required. The basic effort is critical for increasing farmers' knowledge.

ABSTRACT

Background and Aim: Diphenhydramine is an H1-antihistamine that counteracts the toxic effects of organophosphate and carbamate insecticides that inhibit cholinesterase (ChE) activity. This meta-analysis aimed to investigate the effects of diphenhydramine on ChE inhibition induced by these insecticides in the plasma, erythrocytes, or whole brain of experimental animals.

Materials and Methods: A data search was performed on erythrocyte, plasma, and brain ChE inhibition caused by organophosphate and carbamate insecticides in experimental animals (mice, rats, and chicks) treated with the antihistamine diphenhydramine in accordance with preferred reporting items for systematic reviews and meta-analysis, which was done by the two-group random-effects model meta-analysis. The meta-analysis included 18 records extracted from six studies that appeared from 1996 to 2022.

Results: Using the random-effects model, a two-group meta-analysis revealed that the combined effect size (ChE inhibition) was significantly more favorable in the control group than in the diphenhydramine intervention, as shown by a forest plot. The combined effect size (standardized mean difference) was 0.67, with a standard error of 0.3, a lower limit of 0.04, and an upper limit of 1.29 (p = 0.025). The heterogeneity was moderate, as I2 of the combined effect size was 74%, with a significant Cochrane Q-test result (Q = 65, p < 0.0001). Subgroup analysis indicated that, with brain ChE inhibition, the heterogeneity (I2) became 5%, which was lower than ChE inhibition in plasma (84%) and erythrocytes (78%). No publication bias was identified using the funnel plot and Egger's test.

Conclusion: This meta-analysis suggests that, in addition to its documented antidotal action against ChE-inhibiting insecticides, diphenhydramine can also reduce the extent of ChE inhibition, especially in the brain, which is the main site of toxicity of these insecticides. There is a need for additional studies to assess such enzyme inhibition in different parts of the brain.
